Design Notes

Even though this LacI gene is adapted from a yeast optimized version, we decided that since the LacI gene is typically from bacteria, it would be best to run it through a yeast optimizer to double check that it is certainly optimized. To do this, we simply ran the LacI gene from the BBa_K642001 part through the IDT Codon Optimization application. In addition, we added a stop codon (TAA) to the end of the original BBa_K642001 part.

This part was inserted into DNA2.0 Yeast Expression Vector pD1224 (see references).
